目的探讨青黛复方对K-562细胞调亡、Bcr/Abl融合基因及p210蛋白的影响。方法以不同浓度青黛复方(0、2.5、5、7.5、10和20mg/ml)处理K-562细胞24h后,采用半定量RT-PCR技术检测各组Bcr/Abl融合基因的表达,Western blot技术检测p210Bcr/Abl蛋白表达,流式细胞术检测细胞凋亡率。结果随药物作用浓度升高,K-562细胞凋亡率呈浓度依赖性增加;Bcr/Abl在mRNA和蛋白水平均呈浓度依赖性下调,尤以10mg/ml和20mg/ml处理组更为显著。结论青黛复方能部分抑制K-562细胞Bcr/Abl的表达并呈浓度依赖关系,其对慢性髓细胞性白血病(CML)的疗效可能是通过促进细胞凋亡而实现的。
Objective To investigate the effect of Indigo compound on the apoptosis of K-562 cells, Bcr / Abl fusion gene and p210 protein. Methods After treated with different concentrations of indigo compound (0, 2.5, 5, 7.5, 10 and 20 mg / ml) for 24 hours, the expression of Bcr / Abl fusion gene in each group was detected by semi-quantitative RT-PCR. Western blot The expression of p210Bcr / Abl protein was detected, and the apoptosis rate was detected by flow cytometry. Results The apoptotic rate of K-562 cells increased in a concentration-dependent manner with increasing concentration of the drug. Bcr / Abl was down-regulated in mRNA and protein levels in a concentration-dependent manner, especially in the 10 mg / ml and 20 mg / ml groups . Conclusion The compound of Qingdai can partially inhibit the expression of Bcr / Abl in K-562 cells in a concentration-dependent manner, and its therapeutic effect on chronic myelogenous leukemia (CML) may be through the promotion of apoptosis.
