目的研究局部应用血管内皮生长因子(vasular endothelial growth foutor,VEGF)防治肢体严重创伤后局部骨质疏松的作用,为临床应用提供实验依据。方法参照Allen氏垂直重量打击法(weight-dropping,WD法)制作新西兰家兔桡骨远端严重损伤动物模型。将实验动物随机分为实验组(局部注射pHVEGF165质粒)与对照组(局部应用生理盐水组),每组各15只。实验组于伤后即刻局部(桡骨远端周围)注射pHVEGF165质粒50 ug,对照组局部注射生理盐水。于给药后2、4、6、8周观察桡骨远端血流量及骨密度变化。治疗后8周处死动物,行病理学检查。结果实验组伤后2、4、6、8周血流量优于对照组,桡骨远端骨密度明显高于对照组(P<0.05)。实验组病理学检查见骨小梁增多、增粗。结论局部注射pHVEGF165质粒可以改善肢体严重创伤后局部血流量,预防局部骨质疏松的发生。
Objective To study the effect of topical application of vasular endothelial growth factor (VEGF) on the prevention and treatment of local post-traumatic local osteoporosis and provide experimental basis for its clinical application. Methods According to Allen’s weight-dropping (WD) method, a rabbit model of severe distal radius injury in New Zealand rabbits was made. The experimental animals were randomly divided into experimental group (local injection of pHVEGF165 plasmid) and control group (topical saline group), 15 rats in each group. The experimental group was injected with 50 μg of pHVEGF165 plasmid locally (distal to the radius) immediately after the injury, and the control group was injected with normal saline. At 2, 4, 6 and 8 weeks after administration, the changes of distal radius blood flow and bone mineral density were observed. Animals were sacrificed 8 weeks after the treatment and pathological examination was performed. Results The blood flow of the experimental group was better than that of the control group at 2, 4, 6 and 8 weeks after injury, and the distal radius of the distal radius was significantly higher than that of the control group (P <0.05). Experimental group pathological examination showed increased trabecular, thickening. Conclusion Local injection of pHVEGF165 plasmid can improve local blood flow after severe trauma and prevent the occurrence of local osteoporosis.
