该文旨在研究桂枝茯苓胶囊中17个成分的抗炎与免疫调节活性。主要选择LPS刺激的RAW264.7细胞为炎症模型,考察17个成分的体外抗炎作用,ELISA法检测IL-1β,TNF-α,PGE2释放水平、免疫荧光法检测COX-2表达水平;选择脾脏淋巴细胞体外活化增殖为模型,考察17个成分的免疫调节活性,MTT法和Hoechst 33342染色法检测细胞增殖水平。结果显示,在LPS诱导RAW264.7释放炎症因子模型上筛选出10个活性较强成分;在脾脏淋巴细胞活化增殖模型上筛选出10个活性较强成分。剔除重复成分,在上述2个模型上共计筛选出强活性成分15个。以上结果表明,这15个成分可能是桂枝茯苓胶囊抗炎、调节免疫,治疗相关疾病如盆腔炎等的主要活性成分。
The purpose of this paper is to study the anti-inflammatory and immunomodulatory activities of 17 components in Guizhi Fuling capsule. LPS-stimulated RAW264.7 cells were selected as the inflammatory model to investigate the antiinflammatory effects of 17 components in vitro. The levels of IL-1β, TNF-α and PGE2 were detected by ELISA and the expression of COX-2 by immunofluorescence. Lymphocytes in vitro activation and proliferation as a model to examine the immunomodulatory activity of 17 components, MTT assay and Hoechst 33342 staining to detect cell proliferation. The results showed that 10 active components were screened by LPS-induced RAW264.7 release inflammatory cytokines, and 10 active components were screened by spleen lymphocyte activation and proliferation model. Excluding repetitive components, a total of 15 strong active components were screened on the above two models. The above results show that these 15 components may be Guizhifuling capsule anti-inflammatory, immune regulation, treatment-related diseases such as pelvic inflammatory disease, the main active ingredient.
