目的:考察大孔树脂富集牡丹皮中芍药苷的工艺参数。方法:以芍药苷为指标,HPLC测定指标成分含量,采用静态、动态吸附-解吸试验考察大孔树脂型号并优选其富集纯化工艺条件。结果:采用AB-8型大孔树脂,上样液质量浓度0.125 g.mL-1,上样量2.25 BV,上样液pH 4,吸附流速2 BV.h-1,8 BV水洗除杂,14 BV 20%乙醇洗脱芍药苷,洗脱流速4BV.h-1。此条件下,芍药苷洗脱率达90.40%。结论:该优选工艺可较好的富集牡丹皮中有效成分芍药苷,可推广于工业化大生产。
Objective: To investigate the technological parameters of paeoniflorin in the moutan bark enriched by macroporous resin. Methods: Paeoniflorin was used as an index to determine the content of indicator components by HPLC. The static and dynamic adsorption-desorption tests were used to investigate the macroporous resin model and its enrichment and purification conditions were optimized. Results: Using AB-8 macroporous resin, the sample liquid mass concentration of 0.125 g.mL-1, the sample volume of 2.25 BV, the sample liquid pH 4, the adsorption flow rate of 2 BV.h-1, Paeoniflorin was eluted with 14 BV 20% ethanol and the elution flow rate was 4BV.h-1. Under these conditions, paeoniflorin elution rate of 90.40%. Conclusion: The optimal process can enrich the active ingredient paeoniflorin in the peony bark, which can be widely used in industrialized large-scale production.
