目的:建立固相萃取结合高效液相色谱法测定人全血中的环孢素A(CsA)含量,为CsA的血药浓度监测和药动学研究提供测定方法。方法:全血经固相萃取法处理,紫外检测,内标法定量。结果:CsA在50~500ng·ml~(-1)范围内线性关系良好(r=0.999 3)。高、中、低浓度的提取回收率均大于85%,日内、日间RSD均低于10%。结论:该法操作简便,灵敏度高,是一种较为理想的测量方法。
OBJECTIVE: To establish a method for the determination of cyclosporin A (CsA) in human whole blood by solid-phase extraction combined with high performance liquid chromatography (HPLC), and to provide a method for the determination of CsA concentration and pharmacokinetics. Methods: Whole blood was processed by SPE, UV detection, internal standard method. Results: CsA showed a good linearity (r = 0.999 3) in the range of 50 ~ 500ng · ml ~ (-1). The recovery rates of high, medium and low concentrations were all above 85%, and the intra-day and interday RSDs were less than 10%. Conclusion: The method is simple, high sensitivity, is an ideal measurement method.
