血脑屏障(BBB)的基本结构是脑毛细血管内皮细胞间的紧密连接。这方面的研究较多,具有重要意义。本文概要综述关于BBB与BBB开放的研究简况。 一、BBB的概念过去认为,BBB系脉络丛、脑毛细血管壁与脑组织之间的结构,包括血管内皮细胞、基膜、神经胶质细胞突起。近十多年来,由于电子显微镜和生物酶标技术的进展,对BBB及其部位所在提供了重要资料,BBB的概念也远远超出原先的含义。将辣根过氧化酶(HRP)、铁蛋白等示踪剂注入血管后,可见其受阻于内皮细胞问的紧密连接处,不能进入脑组织。如果用另一种方式将上述示踪剂直接注入脑组织或脑脊液中,发现它们不是在神经胶质细胞突起或基膜处受阻,也是被毛细血管内皮细胞及其紧密连接所阻挡,不能进入毛细血管腔,上述结果提示脑毛细血管内皮细胞及其间的紧密连接是BBB的基本结构。而神经胶质细胞突起和基膜并非真正的通透性屏障,它们可能对BBB的通透性起一定辅助作用。脑毛细血管与一般的毛细血管相比有所不同,内皮细胞无窗孔,缺乏吞饮小泡,但细胞间有紧密连
