目的:建立测定胰蛋白酶中有机溶剂丙酮残留量的方法。方法:采用毛细管气相色谱法,色谱柱为Supelco 25301-U型石英毛细管柱;检测器为氢火焰离子化检测器;载气为高纯度氮气;水为溶解介质,检测了胰蛋白酶中丙酮的残留量。结果:丙酮在样品中达到完全分离,在1.59~25.50ppm浓度范围内具有良好的线性,线性方程为A=306.0C+184.3,r= 0.999 5,检测限为0.03ppm,方法回收率为99.9%,RSD为1.7%。结论:本法能快速准确的检测胰蛋白酶中有机溶剂丙酮残留量,可用于胰蛋白酶质量控制中的有机残留溶剂检查。
Objective: To establish a method for determination of acetone residues in organic solvent of trypsin. METHODS: Capillary gas chromatography was performed on a Supelco 25301-U quartz capillary column. The detector was a hydrogen flame ionization detector. The carrier gas was high purity nitrogen. Water was used as the dissolution medium to detect the residual acetone in trypsin the amount. Results: Acetone was completely separated in the sample with a good linearity in the range of 1.59-25.50ppm. The linear equation was A = 306.0C + 184.3, r = 0.999 5, the detection limit was 0.03ppm, the recovery rate was 99.9% , RSD was 1.7%. Conclusion: This method can quickly and accurately detect acetone residues of organic solvent in trypsin and can be used for organic residual solvent examination in the quality control of trypsin.
