目的:探讨二甲双胍对子宫内膜癌(EC)细胞系HEC-1A和Ishikawa中脂联素表达水平的影响,及其可能的作用机理。方法:采用CCK8法检测二甲双胍对EC细胞系HEC-1A和Ishikawa的增殖抑制率。依据CCK8的结果按不同浓度二甲双胍处理HEC-1A和Ishikawa后,运用RT-PCR检测脂联素mRNA的变化、Western Bloting法检测不同浓度二甲双胍作用后FOXO1和脂联素表达水平的变化。结果:二甲双胍能增加脂联素mRNA和蛋白的表达水平,同时提高FOXO1的含量,且呈剂量依赖关系。结论:二甲双胍能提高EC细胞中脂联素的表达,其作用机制可能与FOXO1上调有关。
Objective: To investigate the effect of metformin on the expression of adiponectin in endometrial carcinoma (EC) cell lines HEC-1A and Ishikawa and its possible mechanism. Methods: The inhibitory rate of metformin on the proliferation of EC cell lines HEC-1A and Ishikawa was detected by CCK8 assay. According to the results of CCK8, HEC-1A and Ishikawa were treated with different concentrations of metformin. The mRNA expression of adiponectin was detected by RT-PCR. The expressions of FOXO1 and adiponectin were detected by Western Bloting. Results: Metformin increased adiponectin mRNA and protein expression levels, while increasing FOXO1 content, and in a dose-dependent manner. CONCLUSION: Metformin can increase the expression of adiponectin in EC cells, which may be related to the up-regulation of FOXO1.
