【目的】探讨黄芩苷对脂多糖(LPS)诱导的IEC-6细胞凋亡及迁移能力的影响。【方法】以LPS诱导IEC-6细胞建立细胞损伤模型,采用四甲基偶唑盐(MTT)法和酶联免疫吸附(ELISA)法试剂盒分别检测LPS对IEC-6增殖及分泌炎症因子肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)的影响;应用Hoechst 33258染色和流式细胞术观察和分析黄芩苷低、中、高剂量组(剂量分别为2.5、5.0、10.0μg/m L)对LPS诱导的IEC-6细胞核凋亡形态学改变及凋亡率的影响;应用细胞划痕实验检测黄芩苷对LPS诱导的IEC-6细胞迁移能力的影响。【结果】与对照组比较,1.0μg/m L LPS组IEC-6细胞生存率显著降低,TNF-α、IL-6分泌量及细胞凋亡率显著增加,细胞迁移能力显著下降(均P<0.05);与模型组比较,黄芩苷低、中、高剂量组凋亡细胞均不同程度减少,高剂量组细胞凋亡率显著降低(P<0.05),细胞迁移显著提高(均P<0.01)。【结论】黄芩苷可抑制LPS诱导的肠道上皮细胞的凋亡,并能提高其迁移能力。
【Objective】 To investigate the effects of baicalin on the apoptosis and migration of IEC-6 cells induced by lipopolysaccharide (LPS). 【Methods】 Cell injury models were established by IEC-6 cells induced by LPS. Tetramethylpyrazole (MTT) and enzyme-linked immunosorbent assay (ELISA) were used to detect the effect of LPS on IEC-6 proliferation and secretion of inflammatory cytokines (TNF-α) and interleukin-6 (IL-6). Hoechst 33258 staining and flow cytometry were used to observe and analyze the effects of baicalin at low, medium and high doses (2.5, 5.0,10.0μg / m L) on the morphological changes and apoptotic rate of IEC-6cell induced by LPS. The cell scratch assay was used to detect the effect of baicalin on LPS-induced migration of IEC-6cell. 【Results】 Compared with the control group, the survival rate of IEC-6 cells was significantly decreased and the secretion of TNF-α and IL-6 and the apoptosis rate of IEC-6 cells were significantly increased (P < 0.05). Compared with the model group, the apoptotic cells of baicalin in low, medium and high dose groups were all decreased to a lesser extent, the apoptosis rate of high dose group was significantly decreased (P <0.05), and cell migration was significantly increased (all P <0.01) . 【Conclusion】 Baicalin can inhibit LPS-induced intestinal epithelial cell apoptosis and increase its migration ability.
