目的 探讨同源异型盒DLX4基因在乳腺癌发生、发展进程中的可能作用。方法 采用实时荧光定量PCR技术检测 82例乳腺肿瘤组织及 2 2例癌旁正常腺体中DLX4mRNA的表达情况。结果 82例肿瘤组织中 ,有 5 3例呈DLX4阳性表达 (6 4 6 3% ) ,而在癌旁正常腺体中则表达很弱(4 2 2 ,18 18% ) ,差异具有显著性 (P <0 0 5 )。DLX4基因的过表达与病理组织学分级有关 (P <0 0 5 ) ,而与癌肿大小、淋巴结转移、病理类型、肿瘤家族史、雌激素受体及孕激素受体状态无关。不同病理组织分级的乳腺癌标本中DLX4基因的初始拷贝数分别为 (5 13± 0 90 )× 10 3,(1 4 1± 0 4 8)× 10 4 和(3 0 1± 0 5 3)× 10 4 拷贝 μgRNA ,随着组织学分级的升高 ,DLX4基因mRNA表达量呈上升趋势 (F =8 4 7,P <0 0 5 )。结论 DLX4基因在乳腺肿瘤中呈过度表达 ,可能在某种程度上参与了乳腺癌变的发生和发展。
Objective To investigate the possible role of homeobox DLX4 gene in the development and progression of breast cancer. Methods The expression of DLX4 mRNA in 82 breast tumor tissues and 22 normal adjacent glands was detected by real-time fluorescence quantitative PCR. Results Of the 82 tumor tissues, 53 cases were positive for DLX4 expression (6 4 63%), while in normal paracancerous glands the expression was weak (4 2 2 , 18 18%). The difference was significant ( P <0 0 5 ). The overexpression of DLX4 gene was associated with histopathological grade (P < 0.05), but not with the size of cancer, lymph node metastasis, pathological type, family history of tumor, estrogen receptor and progesterone receptor status. The initial copy number of DLX4 gene in breast cancer specimens with different pathological grades was (5 13 ± 0 90) × 10 3, (1 4 1 ± 0 4 8) × 10 4 and (3 0 1 ± 0 5 3), respectively. × 10 4 copies of μgRNA, with the increase of histological grade, DLX4 mRNA expression showed an upward trend (F = 874, P <0 05). Conclusion DLX4 gene is overexpressed in mammary tumors and may be involved in the occurrence and development of breast cancer to some extent.
