目的:建立用超高效液相色谱(UPLC)法快速测定苏南山肚痛丸中芍药苷含量。方法:采用Acquity BEH C18(2.1 mm×50 mm,1.7μm)色谱柱,以乙腈-0.1%磷酸(15:85)为流动相,流速0.21 mL.min-1,检测波长230 nm,柱温25℃,进样体积2μL,采集频率20 Hz。利用Acquity Waters system进行含量测定。结果:样品中芍药苷在4.5 min内完全分离并对其进行了含量测定,芍药苷进样量在0.026~0.264μg范围内与峰面积呈良好的线性关系(r=0.9999),平均回收率(n=6)为102.2%(RSD=0.4%)。结论:UPLC法在不影响分离效果的情况下大大提高了芍药苷分析速度,同时减少了溶剂消耗。该研究表明UPLC法可作为传统HPLC法的一种更为方便、可行的替代方法。
Objective: To establish a rapid determination of paeoniflorin in Sunan Shantao Wan by ultra performance liquid chromatography (UPLC). METHODS: The mobile phase was acetonitrile-0.1% phosphoric acid (15:85) with a flow rate of 0.21 mL · min-1 on an Acquity BEH C18 column (2.1 mm × 50 mm, 1.7 μm) ℃, injection volume 2μL, acquisition frequency 20 Hz. The content was determined using an Acquity Waters system. Results: Paeoniflorin in the sample was completely separated within 4.5 min and the content of paeoniflorin was determined in the range of 0.026-0.264 μg with a good linear relationship (r = 0.9999). The average recovery ( n = 6) was 102.2% (RSD = 0.4%). Conclusion: The UPLC method greatly improves the speed of paeoniflorin analysis while reducing the solvent consumption without affecting the separation efficiency. This study shows that the UPLC method can be used as a more convenient and feasible alternative method for the traditional HPLC method.
