观察NKx2.5、GATA-4基因在骨髓间充质干细胞 (mesenchymalstemcells,MSCs)分化为心肌细胞过程中表达时序性及表达强度的变化 ,初步阐明两基因对MSCs分化为心肌细胞的影响。以MSCs植入后的Wistar大鼠为研究对象 ,在移植后不同的时间点取心肌标本 ,采用逆转录聚合酶链反应技术 ,提取心肌组织的总RNA ,逆转录 ,加入特异性引物扩增目的基因 ,观察基因表达的时序性 ,并应用凝胶图像分析系统检测这两个基因表达强度的动态变化。显示在MSCs移植后 1d两个基因即出现弱的表达 ,1d~ 1周表达较低 ,2周~ 3周表达强度最高 ,达到高峰期 ,以后表达强度则逐渐降低。NKx2.5、GATA 4在MSCs转化为心肌细胞的早期发挥重要的作用
To observe the changes of the expression sequence and the intensity of expression of NKx2.5 and GATA-4 genes during the differentiation of mesenchymal stem cells (MSCs) into cardiomyocytes, and to clarify the effects of these two genes on the differentiation of MSCs into cardiomyocytes. The Wistar rats implanted with MSCs were used as the research objects. The myocardial samples were collected at different time points after transplantation. The total RNA was extracted from the myocardium by reverse transcription polymerase chain reaction (RT-PCR) Gene, observe the temporal sequence of gene expression, and use the gel image analysis system to detect the dynamic changes of the expression intensity of these two genes. The results showed that the two genes expressed weakly on the 1st day after MSCs transplantation. The expression was low in the first week and the highest in the second week to the third week, reaching the peak and decreasing gradually afterwards.
