用3.5L和14L生物反应器培养WUT3杂交瘤细胞,结果表明用人血清可以代替牛血清培养WuT3细胞,在1%人血清培养液中添加适当营养成分,可达到10%小牛血清浓度下的细胞浓度(1.5×106/ml)和单抗产量(大于50μg/ml)。放大到75L生物反应器中采用半连续方式培养,建立了中试培养工艺流程和质控点,收获时细胞浓度为1.0×106~1.5×106/ml,单抗产量为40~70μg/ml,平均日产单抗达1g。在3.5L、14L、75L逐级放大培养过程中,细胞支原体为阴性,收获上清中单抗免疫活性合格,热原试验合格。
The WUT3 hybridoma cells were cultured in 3.5L and 14L bioreactor.
