目的探讨热休克蛋白60(Hsp60)基因多态性和抗体水平及其与冠心病(CHD)的关系。方法采用病例对照研究,选取未治疗过的CHD患者615例,对照以年龄(±1)岁和性别以1∶1与CHD组匹配共选取615例。采用间接酶联免疫吸附法测定血浆Hsp60抗体水平;TaqM an-MGB探针检测hsp60基因上rs2340690、rs788016、rs2565163和rs2305560四个单核苷酸位点的多态性(SNPs)。结果CHD组的Hsp60抗体水平显著高于对照组,差异有统计学意义(P=0.000)。上述4个SNPs的基因型频率在CHD组和对照组之间差异均无统计学意义,采用多元Lo-gistic回归分析,对年龄、性别、吸烟、饮酒、高血压史、糖尿病史和家族CHD史等因素进行校正后,未发现4个SNPs的任一基因型与CHD的危险度有关联;多因素协方差分析校正了以上混杂因素后,CHD组和对照组hsp60基因4个SNPs不同基因型之间的血浆Hsp60抗体水平差异均无统计学意义(P>0.05)。结论Hsp60抗体可能与CHD发病有关,hsp60基因的多态性和抗体水平与CHD风险均没有显著性关系。
Objective To investigate the relationship between Hsp60 gene polymorphisms and antibody levels and coronary heart disease (CHD). Methods A case-control study was conducted in 615 untreated patients with CHD, and 615 controls were selected according to age (± 1) years and sex matched with CHD group by 1: 1. Plasma Hsp60 antibody levels were determined by indirect enzyme-linked immunosorbent assay. The four SNPs of rs2340690, rs788016, rs2565163 and rs2305560 on hsp60 gene were detected by TaqM an-MGB probe. Results The level of Hsp60 antibody in CHD group was significantly higher than that in control group (P = 0.000). The genotype frequency of the above four SNPs was not significantly different between the CHD group and the control group. Multiplex Lo-gistic regression analysis was used to analyze the age, sex, smoking, drinking, history of hypertension, history of diabetes and family history of CHD After adjusting for these factors, no association was found between the four genotypes of SNPs and the risk of CHD. Multivariate analysis of covariance adjusted for the four genotypes of hsp60 gene in CHD group and control group Between the plasma Hsp60 antibody levels were no significant difference (P> 0.05). Conclusion The Hsp60 antibody may be related to the pathogenesis of CHD. The polymorphism of hsp60 gene and antibody level have no significant relationship with the risk of CHD.
