目的探讨组织因子(TF)表达对大肠癌细胞血行转移能力的影响。方法利用构建有正义/反义组织因子cDNA(TFcDNA)的真核表达载体pcDNA 3.1/Zeo,以脂质体法转染LoVo 细胞,经验证转染成功的LoVo细胞采用Western blot检测组织因子的蛋白表达水平,再将24只裸鼠(BALB/C nu/nu)随机分成3组,分别将转染了正义/反义TFcDNA的LoVo细胞及未转染的Lo- Vo细胞通过脾接种至裸鼠体内。6周后观察裸鼠发生肝表面转移灶的数目,以肝表面转移灶的数目反映LoVo细胞的转移能力。结果与LoVo细胞相比,转染了正义TFcDNA的LoVo细胞的 TF表达水平升高,而转染了反义TFcDNA的LoVo细胞的TF表达水平则降低(F=33.9,P< 0.01)。转染了正义TFcDNA的LoVo细胞组比LoVo组的肝转移灶的数目多,而转染了反义TFcD- NA的LoVo细胞组则比LoVo组的肝转移灶的数目少(F=52.1,P<0.01)。结论组织因子可以增加人类大肠癌细胞的体内血行转移能力。
Objective To investigate the effect of tissue factor (TF) on the hematogenous metastasis of colorectal cancer cells. Methods LoVo cells were transfected into LoVo cells by using eukaryotic expression vector pcDNA3.1 / Zeo constructed with sense / antisense tissue factor cDNA (TFcDNA). The LoVo cells transfected with LoVo cells were verified by Western blot to detect the expression of tissue factor 24 nude mice (BALB / C nu / nu) were randomly divided into 3 groups. LoVo cells transfected with sense / antisense TF cDNA and untransfected Lo-Vo cells were inoculated into nude mice in vivo. Six weeks later, the number of liver metastases in nude mice was observed, and the number of hepatic metastases was counted to reflect the metastasis ability of LoVo cells. Results Compared with LoVo cells, TF expression in LoVo cells transfected with sense TFcDNA increased and that in LoVo cells transfected with antisense TF cDNA decreased (F = 33.9, P <0.01) ). LoVo cells transfected with sense TF cDNAs had more hepatic metastases than LoVo cells, whereas LoVo cells transfected with antisense TFcD-NA had fewer hepatic metastases than LoVo cells (F = 52.1 , P <0.01). Conclusion Tissue factor can increase the ability of human colorectal cancer cells to hematopoietic metastasis.
