以老芒麦×紫芒披碱草种间杂种F1代的幼穗(2~4cm)为外植体材料,以MS为基本培养基,研究了4种不同外源激素组合对幼穗愈伤组织诱导和植株再生能力的影响,筛选出最适杂种F1愈伤组织诱导的培养基为:MS+2,4-D2.0mg/L,诱导率达63.3%;适宜幼苗分化的最佳培养基为:MS+6-BA2.0mg/L+NAA0.4mg/L,分化率为75%;适宜生根的最佳培养基为MS+IBA0.5mg/L,生根率达90%;组培苗经过炼苗移栽得到了完整植株。该再生体系的建立为下一步进行秋水仙碱诱导染色体加倍提供基础,最终克服其不育性,创造饲草新种质。
