目的:探讨Notch-1在甲状腺乳头状癌中的表达特点及其生物学意义。方法:应用免疫荧光法和RT-PCR法,分别检测人甲状腺乳头状癌组织标本中,Notch-1基因和蛋白的表达情况。结果:RT-PCR检测,甲状腺乳头状癌组织中Notch-1mRNA的表达明显低于正常甲状腺组织;免疫荧光法测定,Notch-1在正常甲状腺组织中表达呈强阳性,甲状腺乳头状癌组织中呈弱阳性。Notch-1基因及蛋白表达在甲状腺乳头状癌组织和正常甲状腺组织之间有明显差别(P<0.001)。结论:在甲状腺乳头状癌组织中,Notch-1在mRNA及蛋白表达水平均低调。提示notch-1信号通路受到普遍抑制,该基因在甲状腺癌发生、发展中起抑癌作用。
Objective: To investigate the expression of Notch-1 in papillary thyroid carcinoma and its biological significance. Methods: Immunofluorescence and RT-PCR were used to detect the expression of Notch-1 gene and protein in human thyroid papillary carcinoma tissues. Results: The expression of Notch-1 mRNA in thyroid papillary carcinoma tissues was significantly lower than that in normal thyroid tissues by RT-PCR. The expression of Notch-1 in normal thyroid tissue was strongly positive by immunofluorescence assay. The expression of Notch- Weak positive. Notch-1 gene and protein expression in thyroid papillary carcinoma and normal thyroid tissue were significantly different (P <0.001). Conclusion: The expression of Notch-1 mRNA and protein in thyroid papillary carcinoma is low. Suggesting that notch-1 signaling pathway is generally inhibited, the gene plays a role in tumorigenesis and development of thyroid cancer.
