提取金氏真蛇尾皂苷并研究其抗氧化和抑菌活性,为蛇尾纲动物资源的研究和开发利用提供理论依据。以金氏真蛇尾为原材料,通过乙醇提取、石油醚脱脂、正丁醇萃取及大孔树脂AB-8除杂,制得蛇尾皂苷,其熔点为241~256℃;通过羟自由基法和邻苯三酚自氧化法来测定不同浓度的皂苷溶液对不同自由基的清除能力,采用滤纸片法及最低抑菌浓度(MIC)法测定金氏真蛇尾皂苷对大肠杆菌、金黄色葡萄球菌、啤酒酵母菌和青霉的抑制作用。金氏真蛇尾皂苷对·OH和O2-具有较好的清除作用,对大肠杆菌、金黄色葡萄球菌和啤酒酵母菌具有较好的抑制作用,对青霉的抑制作用较弱。通过抗氧化和抑菌活性的研究,表明金氏真蛇尾皂苷具有较好的抗氧化及抑菌活性。
Extraction of Geniposide and study its anti-oxidation and antibacterial activity, provide a theoretical basis for the research and development and utilization of resources. The results showed that the average molecular weight of the polysaccharides was about 90% .With the Herba Juvenile Hemsleyi as the raw material, the ethanolic extract, petroleum ether degreasing, n-butanol extraction and macroporous resin AB-8 were used for impurity removal. Pyrogallol autoxidation method was used to determine the scavenging ability of different concentrations of saponin solution to different free radicals. The filter paper method and the minimum inhibitory concentration (MIC) method were used to determine the effect of Genipostella saponin on Escherichia coli, Staphylococcus aureus, beer Yeast and Penicillium inhibition. Geniposide has good scavenging effect on · OH and O2-, has good inhibitory effect on Escherichia coli, Staphylococcus aureus and Saccharomyces cerevisiae, and has a weak inhibitory effect on Penicillium. Antioxidant and antibacterial activities of the research shows that Geniposide has good anti-oxidation and antibacterial activity.
