目的:研究天麻素对过氧化氢损伤大鼠原代皮层神经元的保护作用。方法:本研究采用过氧化氢损伤大鼠皮层神经元模型,在造模同时给予不同浓度的天麻素溶液,采用DPPH法测定天麻素对氧自由基的清除能力;通过检测尿酸生成,测定天麻素对过氧化氢损伤后大鼠皮层神经元中黄嘌呤氧化酶活性的影响。提取蛋白,通过WB法检测BDNF表达。结果:天麻素能够逆转过氧化氢对大鼠原代皮层神经元造成的损伤,降低细胞上清液中LDH含量,提高活细胞量;细胞上清中SOD活性升高;天麻素可清除氧自由基,抑制黄嘌呤氧化酶活性;天麻素能够促进损伤神经元中BDNF的表达。结论:天麻素对过氧化氢损伤的神经元有显著保护作用。
Objective: To investigate the protective effects of gastrodin on primary cortical neurons injured by hydrogen peroxide in rats. Methods: In this study, cortical neurons were induced by hydrogen peroxide (H 2 O 2) injury in rats. Gastrodin solution with different concentrations was given at the same time. The DPPH method was used to determine the scavenging capacity of gastrodin to oxygen free radicals. By detecting the formation of uric acid, Effects of Xanthine Oxidase Activity in Rat Cortical Neurons after Hydrogen Peroxide Injury. Protein was extracted and BDNF expression was detected by WB method. Results: Gastrodin could reverse the damage caused by hydrogen peroxide to the primary cortical neurons in rats, reduce the content of LDH in the supernatant and increase the amount of viable cells. The activity of SOD in the supernatant of the gastrodin increased. Gastrodin cleared the oxygen free , Inhibit xanthine oxidase activity; Gastrodin can promote the expression of BDNF in injured neurons. Conclusion: Gastrodin has a significant protective effect on hydrogen peroxide injured neurons.
