目的:通过观察通脉宁颗粒(TMN)对自发性高血压大鼠(SHR)血压及胸主动脉血管重构的影响,为该药临床推广提供实验依据。方法:以 SHR 为模型,采用无创性套尾法测定尾动脉收缩压,观察TMN 对 SHR 血压的影响,并结合电镜及图像分析观察 TMN 对 SHR 胸主动脉血管重构的影响。结果:给药4周时,给药组的血压明显低于空白对照组(P<0.01);给药组与正常对照组的血管中层纤维成分体密度(Vv1)和血管平滑肌细胞(VSMC)线粒体(Vv2)均小于空白对照组(P<0.01);给药组与正常对照组 VSMC线粒体的表面均大于空白对照组(P<0.01)。结论:TMN 对 SHR 有降低血压、逆转胸主动脉血管重构的作用。
Objective: To observe the effect of Tongmaining Granules (TMN) on blood pressure and thoracic aortic vascular remodeling in spontaneously hypertensive rats (SHR), and provide experimental evidence for its clinical application. Methods: SHR was used as a model to measure the systolic blood pressure of the caudal artery using the non-invasive tail method. The effect of TMN on blood pressure of SHR was observed. The effects of TMN on vascular remodeling of thoracic aorta in SHR were observed by electron microscopy and image analysis. RESULTS: At 4 weeks after administration, the blood pressure in the administration group was significantly lower than that in the control group (P<0.01); the midstream vascular fiber body density (Vv1) and the vascular smooth muscle cells (VSMC) mitochondria in the administration group and the normal control group. (Vv2) were less than the blank control group (P<0.01); the surface of the mitochondria of the VSMCs in the administration group and the normal control group was larger than the blank control group (P<0.01). Conclusion: TMN can reduce blood pressure and reverse thoracic aortic vascular remodeling in SHR.
