采用高效液相色谱法测定300多种市售主食食材中β-胡萝卜素含量,具体条件:色谱柱为Agilent Zobrax SB-C_(18)(4.6 mm×250 mm,5μm),流动相条件为:含0.4 g/L抗坏血酸的甲醇/乙腈/三氯甲烷(84/12/4)溶液(避光储存);流速1.5 m L/min;柱温:30℃;检测波长:450 nm,进样体积10μL,外标法定量。结果表明该方法在0.500~20.0μg/mL质量浓度范围内线性良好,相关性系数R=0.999 9,RSD=2.4%,方法检出限为0.2 mg/kg,回收率为78.7%~96.4%。该方法整体操作简单,线性范围好,结果可靠。该方法通过高效液相色谱法完成了300多种深圳市售主食材中β-胡萝卜素含量测定,首次建立β-胡萝卜素含量数据库,并与我国现有的胡萝卜素含量数据库和美国β-胡萝卜素含量数据库进行了比较。
The content of β-carotene in more than 300 kinds of commercial staple foods was determined by high performance liquid chromatography. The specific conditions were as follows: Agilent Zobrax SB-C 18 (4.6 mm × 250 mm, 5 μm) Methanol / acetonitrile / trichloromethane (84/12/4) solution containing 0.4 g / L ascorbic acid (stored in dark); flow rate 1.5 m L / min; column temperature 30 ° C .; detection wavelength 450 nm; injection volume 10μL, external standard method. The results showed that the method was linear in the range of 0.500 ~ 20.0μg / mL with a correlation coefficient R = 0.999 9, RSD 2.4%.
