建立了柱前衍生化反相高效液相色谱测定大鼠血浆中的丙二醛(MDA)的方法。以甲基丙二醛(Me-MDA)为内标,2,4-二硝基苯肼(DNPH)进行衍生化反应。采用ZORBAX SB C_(18)柱(250mm×4.6mm,5μm),流动相为0.2%冰醋酸溶液-乙腈(V/V)=50:50,流速为1.0mL/min,检测波长为310nm。丙二醛在2.5—60μmol/L范围内线性关系良好,r=0.9991,平均加标回收率为97.7%,相对标准偏差RSD为4.47%(n=6)。方法准确可靠,专属性好,样品用量少(50μL的血浆),适于大鼠血浆中丙二醛的含量测定。
A method for the determination of malondialdehyde (MDA) in rat plasma by precolumn derivatization reversed-phase high performance liquid chromatography was established. The derivatization of 2,4-dinitrophenylhydrazine (DNPH) with methylmalonaldehyde (Me-MDA) as internal standard. The mobile phase consisted of 0.2% glacial acetic acid-acetonitrile (V / V) = 50:50, the flow rate was 1.0 mL / min, and the detection wavelength was 310 nm using a ZORBAX SB C 18 column (250 mm × 4.6 mm, 5 μm). The linearity of malondialdehyde in the range of 2.5-60μmol / L was good, r = 0.9991, the average recovery was 97.7% and the relative standard deviation (RSD) was 4.47% (n = 6). The method is accurate and reliable, with good specificity and less sample (50μL of plasma), suitable for the determination of the content of malondialdehyde in rat plasma.
