目的研究白纹伊蚊卵在滞育以及冷暴露条件下,热休克蛋白70基因(Hsp70)的表达情况,探究Hsp70在增强白纹伊蚊卵耐寒性方面的作用。方法以无冷暴露的非滞育卵作为对照组,采用RT.qPCR方法检测白纹伊蚊滞育卵和非滞育卵Hsp70 mRNA的表达情况。结果室温25℃条件下,滞育卵与非滞育卵表达的Hsp70差异无统计学意义(P=0.392)。与对照组相比,冷暴露后蚊卵Hsp70表达量均增加,且滞育卵Hsp70的表达量明显高于非滞育卵的表达量,分别是其对照组的16.44和5.22倍。结论Hsp70并不是白纹伊蚊滞育期间的重要组成部分,但是在冷暴露的恢复期表达量上调。
Objective To investigate the expression of heat shock protein 70 (Hsp70) gene in Aedes albopictus eggs under diapause and cold exposure conditions and to explore the role of Hsp70 in enhancing the cold tolerance of Aedes albopictus eggs. Methods Non-diapause eggs without cold exposure were used as control group. RT-qPCR method was used to detect the expression of Hsp70 mRNA in diapause and non-diapause eggs. Results There was no significant difference in Hsp70 expression between diapause and non-diapause eggs at 25 ℃ (P = 0.392). Compared with the control group, the expression levels of Hsp70 in mosquito eggs increased after cold exposure, and the expression levels of Hsp70 in diapause eggs were significantly higher than those in non-diapause eggs, which were 16.44 and 5.22 folds of the control group respectively. Conclusion Hsp70 is not an important part of diapause period of Aedes albopictus, but up-regulated in the convalescent phase of cold exposure.
