目的探讨精索静脉曲张(VC)对青春期大鼠生精细胞HIF-1α、Bcl-2和bax的影响。方法通过部分结扎左肾静脉方法制作左侧精索静脉曲张模型(EV),雄性wistar大鼠36只,随机分为6组,EV持续2周、4周、8周组和相应的接受假手术的对照组(每组6只)。于造模成功后2周、4周、8周取实验组及对照组左侧睾丸,并用免疫组化方法检测睾丸组织HIF-1α、Bcl-2和bax的表达,用TUNEL法检测细胞凋亡并计算生精细胞的凋亡指数(AI)。结果实验组HIF-1α、bax较对照组表达明显升高(P<0.05),而实验组Bcl-2表达较对照组明显下降(P<0.05)。实验组AI较对照组明显升高(P<0.05)。结论精索静脉曲张可致大鼠睾丸组织缺氧,从而诱导睾丸组织细胞凋亡增加,影响睾丸生精功能。
Objective To investigate the effects of varicocele (VC) on spermatogenic cells (HIF-1α, Bcl-2 and bax) in adolescent rats. Methods Left varicocele model (EV) was established by partial ligation of the left renal vein. Thirty-six male Wistar rats were randomly divided into 6 groups. EV was given for 2 weeks, 4 weeks, 8 weeks, and corresponding sham operation Control group (6 in each group). The left testes of experimental group and control group were taken at 2 weeks, 4 weeks and 8 weeks after successful model establishment. The expression of HIF-1α, Bcl-2 and bax in testes was detected by immunohistochemistry and apoptosis was detected by TUNEL The apoptosis index (AI) of spermatogenic cells was calculated. Results The expression of HIF-1α and bax in experimental group was significantly higher than that in control group (P <0.05), while the expression of Bcl-2 in experimental group was significantly lower than that in control group (P <0.05). The experimental group AI was significantly higher than the control group (P <0.05). Conclusions Varicocele can induce hypoxia in testicular tissue of rats and induce apoptosis of testicular tissue and affect the function of testicular spermatogenesis.
