目的探讨妊娠期糖尿病(gestational diabetes mellitus,GDM)与孕妇血清、脐血和胎盘组织中核转录因子NF-κBp65的相关性。方法收集2011年11月至2013年3月天津市中心妇产科医院157名足月妊娠孕妇及正常未孕女性的临床资料,其中GDM孕妇38例(Ⅰ组)、正常孕妇56名(Ⅱ组)、正常未孕女性63名(Ⅲ组)。采用酶联免疫吸附测定3组研究对象血清、脐血中NF-κBp65的表达,采用免疫组化法检测孕妇胎盘组织中NF-κBp65的表达,并进行比较分析。结果Ⅰ组母体血清和脐血中NF-κBp65水平均高于Ⅱ组(均P<0.05);Ⅱ组、Ⅲ组母体血清中NF-κBp65水平比较差异无统计学意义(P>0.05);Ⅰ组胎盘组织中的NF-κBp65表达阳性率100.0%(38/38)高于Ⅱ组的12.5%(7/56)(P<0.05)。结论 NFKBp65可能在GDM的病理生理过程中发挥了一定作用。
Objective To investigate the relationship between gestational diabetes mellitus (GDM) and nuclear factor NF-κBp65 in serum, cord blood and placenta of pregnant women. Methods The clinical data of 157 full-term pregnant women and normal non-pregnant women from Tianjin Central Obstetrics and Gynecology Hospital from November 2011 to March 2013 were collected. Among them, 38 cases of GDM pregnant women (Ⅰ group) and 56 normal pregnant women (Ⅱ group ), 63 non-pregnant normal women (group Ⅲ). Enzyme-linked immunosorbent assay was used to detect the expression of NF-κBp65 in serum and cord blood of three groups. The expression of NF-κBp65 in placenta was detected by immunohistochemistry and compared. Results The levels of NF-κBp65 in serum and cord blood of group Ⅰ were significantly higher than those in group Ⅱ (all P <0.05). There was no significant difference in the serum levels of NF-κBp65 between group Ⅱ and group Ⅲ (P> 0.05) The positive rate of NF-κBp65 in group placenta was 100.0% (38/38), which was higher than that in group Ⅱ (12.5%, 7/56) (P <0.05). Conclusion NFKBp65 may play a role in the pathophysiology of GDM.
