目的:观察一盘珠颗粒对实验性家兔急性软组织损伤后血肿局部软组织NO(一氧化氮)的影响以及局部软组织的形态学改变。方法:建立实验性家兔急性软组织损伤后血肿模型。随机分为一盘珠颗粒高剂量组(5.4 g/kg)、中剂量组(2.7 g/kg)、低剂量组(1.35 g/kg)、三七片组(2.7 g/kg)、正常组、盐水对照组。除正常组外、其余各组分别应用一盘珠颗粒或对照药物或等剂量生理盐水腹腔注射。在治疗后第3天、7天于损伤局部软组织取材,在光镜下观察其组织形态学变化;并将部分组织进行组织匀浆,采用比色法测定匀浆液NO含量。结果:一盘珠治疗组,三七片组损伤局部软组织的淤血明显消退,肌纤维的修复也较盐水组迅速:盐水组损伤局部软组织NO含量在伤后各时相均明显高于正常组(P<0.05);一盘珠颗粒呈剂量依赖方式抑制模兔损伤组织内NO含量的升高(P<0.05),且与三七片组比较,无显著性差异(P>0.05)。结论:一盘珠颗粒能显著促进损伤局部瘀血的消散,并能加快肌纤维的修复。一盘珠颗粒治疗急性损伤性血肿的疗效机理之一与降低局部软组织细胞释放的自由基NO含量有关。
OBJECTIVE: To observe the effect of Yipanzhu Granules on NO (nitrogen oxide) in the soft tissues of local hematoma after acute soft tissue injury in rabbits and the morphological changes of local soft tissues. METHODS: An experimental hematoma model of acute soft tissue injury was established in rabbits. Randomly divided into a high-dose group of Panzhu particles (5.4 g/kg), middle-dose group (2.7 g/kg), low-dose group (1.35 g/kg), Sanqi tablet group (2.7 g/kg), normal group Saline control group. In addition to the normal group, the rest of the groups were administered intraperitoneal injection of a single bead granulate or control drug or an equal dose of saline. On the 3rd and 7th days after treatment, the soft tissue was obtained from the lesions, and the histomorphological changes were observed under a light microscope. Part of the tissue was homogenized and the NO content in the homogenate was measured by a colorimetric method. Results: In the one-bead treatment group, the congestion of the local soft tissue in the Sanqi tablet group significantly subsided, and the repair of muscle fibers was also faster than that in the saline group. The content of NO in the soft tissue in the saline group was significantly higher than that in the normal group at each time after the injury (P <0.05); Yipanzhu Granule inhibited the increase of NO content in the damaged rabbit tissue in a dose-dependent manner (P<0.05), and there was no significant difference compared with the Sanqi tablet group (P>0.05). Conclusion: Yipanzhu granules can significantly promote the dissipation of localized hemorrhoids and accelerate the repair of muscle fibers. One of the therapeutic mechanisms of Yipanzhu granules in the treatment of acute traumatic hematoma is related to the reduction of free radical NO content released by local soft tissue cells.
