目的:对更昔洛韦超声定位靶向释药微囊的制备工艺、含量测定及体外释药行为进行初步研究。方法:用双乳化溶剂挥发法制备微囊,正交试验优化制备工艺,紫外分光光度法测定微囊包封率与载药量,超声诊断仪进行体外超声成像试验,高效液相色谱法测定微囊体外药物释放情况。结果:根据优化工艺制备的更昔洛韦微囊外型圆整,平均粒径为1.28μm,包封率为40.81%,载药量为3.02%,体外成像清晰,未加超声影响时微囊具有良好的缓释作用,在超声影响下微囊可在25min内将药物释放完毕。结论:更昔洛韦超声定位靶向释药微囊具有在超声控制下释放药物的能力,有潜在的应用价值。
OBJECTIVE: To study the preparation technology, content determination and in vitro release behavior of ganciclovir targeted ultrasound-targeted microcapsules. Methods: The microcapsules were prepared by the double emulsion solvent evaporation method, the orthogonal experiment was used to optimize the preparation process, the encapsulation efficiency and drug loading of the microcapsules were determined by ultraviolet spectrophotometry, the ultrasound imaging was performed by the ultrasonic diagnostic apparatus, Capsule drug release outside the body. Results: The ganciclovir microencapsulation was round and round with average particle size of 1.28 μm, encapsulation efficiency of 40.81% and drug loading of 3.02%. The in vitro imaging was clear and the microcapsules Has a good sustained-release effect, under the influence of ultrasound microcapsules within 25min drug release is completed. Conclusion: The ganciclovir-targeted ultrasound-targeted drug-releasing microcapsules have the potential of releasing drugs under the control of ultrasound.
