目的研制一种分散性好、磁响应能力强的高性能Fe_3O_4@SiO_2磁性复合微球,以配合核酸提取仪快速、自动分离纯化DNA。方法通过透射电镜(TEM)、超导量子干涉磁强计(SQUID)对制备的磁性复合微球进行表征。利用紫外分光光度计、实时荧光定量PCR(real time fluorescence quantitative PCR,RQ-PCR)及电泳验证DNA提取效果。结果制备的Fe_3O_4@SiO_2磁性复合微球分散性良好,粒径均匀,且具有良好的超顺磁性和较高的饱和磁化强度。核酸提取实验表明,T100磁性复合微球可从鲑鱼精、乙肝病毒及质粒菌等样品中提取出高质量的DNA。结论制备出一种高性能Fe_3O_4@SiO_2磁性复合微球,可应用于核酸提取仪。
OBJECTIVE To develop a high-performance Fe 3 O 4 @ SiO 2 magnetic composite microspheres with good dispersibility and high magnetic response ability to rapidly and automatically separate and purify DNA with nucleic acid extractor. Methods The prepared magnetic composite microspheres were characterized by transmission electron microscopy (TEM) and superconducting quantum interference magnetometer (SQUID). The DNA extraction efficiency was verified by ultraviolet spectrophotometer, real time fluorescence quantitative PCR (RQ-PCR) and electrophoresis. Results The prepared Fe_3O_4 @ SiO_2 magnetic composite microspheres had good dispersibility, uniform particle size, good superparamagnetism and high saturation magnetization. Nucleic acid extraction experiments showed that, T100 magnetic composite microspheres can be extracted from salmon sperm, hepatitis B virus and plasmid bacteria and other samples of high quality DNA. Conclusion A kind of high performance Fe_3O_4 @ SiO_2 magnetic composite microspheres was prepared and applied to nucleic acid extraction instrument.
