目的研究WCX纳米磁珠联合表面增强激光解析电离化飞行时间质谱(surface-enhanced laser desorption i-onization time-of-flight mass spectrometry,SELDI-TOF-MS)对胃癌及其癌前病变诊断的意义。方法应用纳米磁珠联合SELDI-TOF-MS对26例胃癌、35例癌前病变患者(萎缩性胃炎15例,胃溃疡20例)及30例正常人血清标本进行分析,Swiss蛋白数据库搜索鉴定差异蛋白,建立诊断模型。结果与健康对照相比较,质荷比为2 741.2、7 963.6和15 904.3的蛋白质分子随病变程度表达上调,质荷比为13 732.6的蛋白质在癌前病变组无统计学差异,但在胃癌患者中下调。质荷比2 741.2、7 963.6、15 904.3及13 732.6的蛋白质组成的诊断模型可以准确鉴别胃癌与正常对照,敏感性为92.3%,特异性为96.7%。结论 WCX纳米磁珠联合SELDI-TOF-MS是筛选胃癌早期诊断标志物的有效手段,2 741.2、7 963.6、15 904.3及13 732.6的蛋白质组成的诊断模型有可能成为潜在的胃癌早期诊断标志物。
Objective To investigate the significance of WCX nanospheres coupled with surface-enhanced laser desorption i-onization time-of-flight mass spectrometry (SELDI-TOF-MS) in the diagnosis of gastric cancer and its precancerous lesions. Methods The serum samples from 26 patients with gastric cancer, 35 patients with precancerous lesions (15 patients with atrophic gastritis, 20 patients with gastric ulcer) and 30 normal individuals were analyzed by nano-magnetic beads combined with SELDI-TOF-MS. The differences in the Swiss protein database search Protein, to establish a diagnostic model. Results Compared with healthy controls, the protein molecules with mass-to-charge ratios of 2 741.2, 7 963.6 and 15 904.3 were up-regulated with the degree of lesion. The protein with a mass-to-charge ratio of 13 732.6 had no statistical difference in precancerous lesions but in patients with gastric cancer Down. The diagnosis model of protein with mass-to-charge ratio of 2 741.2, 7 963.6, 15 904.3 and 13 732.6 can accurately identify gastric cancer and normal controls with a sensitivity of 92.3% and a specificity of 96.7%. Conclusion WCX nanobeads combined with SELDI-TOF-MS is an effective method for screening early diagnosis markers of gastric cancer. The diagnostic models of protein composition of 2 741.2, 7 963.6, 15 904.3 and 13 732.6 may be potential markers for early diagnosis of gastric cancer.
