目的 研究核内不均一核糖核蛋白A2 /B1(HnRNPA2 /B1)与肺癌的相关性及其用于肺癌早期诊断的可行性。方法 利用RT -PCR方法分别扩增癌组织、癌旁组织、正常组织、良性病变组织、纤支镜和经皮穿刺肺活检标本中HnRNPA2 /B1外显子 1~ 2 ,分析比较其表达水平的差异及变化趋势。结果 ①HnRNPA2 /B1在正常肺组织、癌旁组织、肺癌组织中表达水平依次增高 ,其阳性率分别为 6 .98%、37.2 %、76 .7%,三组比较有显著性差异 (Ρ <0 .0 5 ) ;HnRNPA2 /B1表达水平在不同的病理类型中的表达阳性率虽有不同 (鳞癌 88.2 %,小细胞肺癌 5 7.1%) ,而且随病理分化程度的降低而增高 ,但是均无统计学差异 (Ρ >0 .0 5 )。②纤支镜和经皮穿刺肺活检标本中HnRNPA2 /B1外显子 1~ 2mRNA阳性率为 6 0 %( 6 /10 ) ,与手术标本的癌组织阳性率比较稍低 ,但无统计学差异(Ρ >0 .0 5 )。结论 HnRNPA2 /B1是肺癌发生、发展过程中的早期事件 ,有可能用于肺癌的早期诊断。
Objective To investigate the correlation between nuclear heterogeneous ribonucleoprotein A2 / B1 (HnRNPA2 / B1) and lung cancer and its feasibility for early diagnosis of lung cancer. Methods The HnRNPA2 / B1 exon 1-2 in lung cancer tissues, adjacent normal tissues, normal tissues, benign lesions, bronchofibroscopy and percutaneous lung biopsy specimens were respectively amplified by RT-PCR. The expression levels of HnRNPA2 / Differences and trends. Results ① The positive expression rates of HnRNPA2 / B1 in normal lung tissue, adjacent non-cancerous tissue and lung cancer tissues were successively higher than those in normal lung tissues (6 .98%, 37.2%, 76.7%, P <0 .0 5). Although the positive expression rates of HnRNPA2 / B1 were different in different pathological types (88.2% in squamous cell carcinoma and 7.11% in small cell lung cancer), the positive rate of HnRNPA2 / B1 increased with the degree of pathological differentiation, but none Statistical difference (Ρ> 0.05). ② The positive rate of HnRNPA2 / B1 exon 1 ~ 2 mRNA in bronchofibroscopy and percutaneous lung biopsy specimens was 60% (6/10), which was slightly lower than that of the surgical specimens, but there was no significant difference (Ρ> 0 .0 5). Conclusion HnRNPA2 / B1 is an early event in the development and progression of lung cancer, which may be used for the early diagnosis of lung cancer.
