目的研究白念珠菌的分子特征,建立分子流行病学研究方法—多位点序列分型技术(MLST),并对临床分离白念珠菌进行分型研究。方法对33株白念珠菌提取DNA,选择7对管家基因进行PCR扩增并将目的基因产物测序,然后将测序结果与标准序列比对后上传至数据库,获得相应的由7对管家基因组成的等位基因谱,最后将等位基因谱再次提交网站确认序列分型(sequence types,STs)。同时与ABC(ATP-binding cassette)分型结果进行比较。结果 33株临床白念珠菌通过MLST产生了33个STs,通过ABC分类为3型。结论 MLST具有较高的分辨力,是一种方便、快速的分子生物学方法,可用于流行病学和菌群多态性的研究。
Objective To study the molecular characteristics of Candida albicans and to establish a molecular epidemiological method - multilocus sequence typing (MLST). The clinical isolates of Candida albicans were genotyped. Methods DNA was extracted from 33 Candida albicans, 7 housekeeping genes were selected for PCR amplification and the target gene products were sequenced. Then, the sequencing results were compared with the standard sequences and uploaded to the database to obtain the corresponding database of 7 pairs of housekeeping genes Allele spectrum, and finally submitted the allelic profile to the website again to confirm sequence types (STs). At the same time with the ABC (ATP-binding cassette) typing results were compared. Results 33 clinical Candida albicans produced 33 STs by MLST and classified as type 3 by ABC. Conclusion MLST has high resolution and is a convenient and rapid molecular biological method for the study of epidemiology and bacterial population polymorphism.
