目的:探讨四逆散对实验性溃疡性结肠炎(UC)大鼠白介素-1β(IL-1β)的影响,以及君臣配伍在组方中的作用。方法:将实验动物分为5组,采用免疫法造模,以大鼠结肠组织大体情况、结肠组织病理变化和IL-1β水平作为观测指标,用ELISA法检测大鼠结肠黏膜组织和血清IL-1β水平。结果:模型组大鼠结肠组织黏膜层可见溃疡形成,且有明显的炎症存在,认为本实验造模成功;模型组大鼠血清和结肠黏膜组织的IL-1β分别为(841.93±95.64)ng·L-1和(75.06±2.15)ng·L-1均显著高于正常组(P<0.01),灌服柴芍枳(1.31 g·kg-1)和柴芍(0.88 g·kg-1)后大鼠血清和结肠黏膜组织IL-1β显著降低(P<0.05),而灌服四逆散(1.75 g·kg-1)后大鼠结肠黏膜组织IL-1β(72.62±2.67)ng·L-1明显减少(P<0.01),但血清IL-1β(1015.52±176.47)ng·L-1却显著升高。结论:四逆散能够通过调节IL-1β来干预实验性UC,其全方发挥作用环节可能为抑制IL-1β由外周血单核细胞向炎症肠黏膜的移行;而方中君、臣药物配伍的作用环节为抑制IL-1β的分泌。
Objective: To investigate the effect of Sini Decoction on interleukin-1β (IL-1β) in experimental ulcerative colitis (UC) rats and the effect of monarch and sage on the prescription. Methods: The experimental animals were divided into five groups, and the immune system was used to make the model. The general condition of the colon, the histopathological changes of the colon and IL-1β level were taken as the observation indexes. The expressions of IL- 1β level. Results: The ulceration of colonic mucosa was observed in the model group, and the inflammation was obvious. The model of the model was successfully established. The levels of IL-1β in the serum and colonic mucosa of model group were (841.93 ± 95.64) ng · L-1 and (75.06 ± 2.15) ng · L-1 were significantly higher than those in normal group (P <0.01) IL-1β in the serum and colonic mucosa of rats decreased significantly (P <0.05), while IL-1β (72.62 ± 2.67) ng · L in colonic mucosa of rats after oral administration of 1.75 g · kg -1 -1 significantly decreased (P <0.01), but serum IL-1β (1015.52 ± 176.47) ng · L-1 was significantly increased. CONCLUSIONS: Sini Decoction can interfere with experimental UC by regulating IL-1β, and its all-round function may inhibit the migration of IL-1β from peripheral blood mononuclear cells to inflammatory intestinal mucosa.
