利用RACE技术首次从药用真菌桦褐孔菌中克隆得到了与菌核形态发育相关的内切葡聚糖编码(EG)基因。结果表明:桦褐孔菌内切葡聚糖酶基因(IO-EG)的cDNA全长为1 315 bp,其中编码区占1 233 bp,具有13个内含子和14个外显子,共编码410个氨基酸。系统进化树分析表明,桦褐孔菌EG与嗜蓝孢孔菌EG在分子进化关系上最相近。实时荧光定量PCR分析表明,随着菌核的生长发育,IO-EG基因的表达量不断升高,初步证明内切葡聚糖酶基因的表达量与桦褐孔菌菌核的产量相关。
The endoglucan encoding (EG) gene related to sclerotial morphology was cloned from the medicinal fungus Inonotus obliquus for the first time using RACE technology. The results showed that the full length cDNA of Io gonorrhoeae endoglucanase gene (IO-EG) was 1 315 bp, encoding 1 233 bp in length with 13 introns and 14 exons Encoding 410 amino acids. Phylogenetic tree analysis showed that the molecular evolution relationships between EG and A. hydrophila EG were the closest. Real-time quantitative PCR analysis showed that with the growth of sclerotia, the expression of IO-EG gene was continuously increased. It was initially proved that the expression of endoglucanase gene was related to the yield of sclerotia.
