目的研究壳聚糖基水凝胶对大鼠肝脏和肾脏的毒性。方法大鼠长期腹腔注射壳聚糖基水凝胶,分别于给药2、4、6周时测定相关血清生化指标(AST、ALT、ALP、TP、ALB、CHO、GLU、BUN、CRE),计算肝、肾脏器系数,并进行肝、肾病理学观察。结果与对照组相比,肾功能相关的BUN指标,给药2周时,低、高剂量组均差异极显著(P<0.01),给药4周时,高剂量组差异显著(P<0.05),其余生化指标均差异不显著(P>0.05);与对照组相比,低、高剂量组肝、肾脏器系数均差异不显著(P>0.05);镜下各组肝脏结构正常,前期低、高剂量组肾脏均有炎性细胞浸润,后期未见明显炎性细胞。结论该壳聚糖基水凝胶对大鼠肝脏未表现出明显毒性作用;对大鼠肾脏前期可引起轻微损伤、后期损伤恢复,无蓄积毒性。
Objective To study the toxicity of chitosan-based hydrogels to rat liver and kidney. Methods Chronic intraperitoneal injection of chitosan-based hydrogels was used to determine the serum levels of biochemical markers (AST, ALT, ALP, TP, ALB, CHO, GLU, BUN and CRE) Calculate the liver, kidney coefficient, and liver, kidney pathology. Conclusion The chitosan-based hydrogel has no obvious toxic effect on the liver of rats. It can cause slight damage to the kidneys in the early stage and restore injury in the late stage without accumulation of toxicity.
