目的:研究六君二陈解毒汤及其拆方对小鼠Lewis肺癌转移的抑制作用及对移植瘤细胞VEGF和nm23-H1表达的影响。方法:建立C57BL/6J小鼠Lewis肺癌模型,给药15 d后观察肺表面转移瘤结节数;用免疫组化法检测Lewis肺癌移植瘤细胞VEGF和nm23-H1的表达。结果:六君二陈解毒汤及其拆方六君二陈汤、清热解毒汤3个治疗组的肺转移形成灶数皆少于生理盐水对照组,其中六君二陈解毒汤肺转移抑制率为80%,明显优于其拆方;六君二陈解毒汤及拆方均可以降低移植瘤细胞VEGF的表达并提高nm23-H1的表达水平。结论:六君二陈解毒汤及其拆方能抑制小鼠Lewis肺癌肿瘤自发性肺转移的发生,其抗肿瘤转移机制可能与影响Lewis肺癌细胞VEGF和nm23-H1的表达有关。
Objective: To study the inhibitory effect of Liujun Er Chen Jie Du Decoction and its decomposed formulas on the metastasis of Lewis lung carcinoma in mice and the effect on the expression of VEGF and nm23-H1 in xenograft tumor cells. Methods: Lewis lung carcinoma model was established in C57BL / 6J mice. The number of metastatic nodules on the surface of lung was observed 15 d after administration. The expression of VEGF and nm23-H1 in Lewis lung carcinoma cells was detected by immunohistochemistry. Results: Liujun Erchen Jiedu Decoction and its decoction Liujun Erchen Decoction, Qingrejiedu Decoction three treatment groups were less than the number of lung metastasis lesion compared with saline control group, including Liujun Erchen decoction soup lung metastasis inhibition rate of 80 %, Which is obviously superior to its disassembled prescription. The effect of Liujun Erchenjiedudu Decoction and its disassembled prescriptions can reduce the expression of VEGF and increase the expression of nm23-H1 in xenografted tumor cells. CONCLUSION: Liujun Erchen Jiedu Decoction and its decomposed formulas can inhibit the spontaneous lung metastasis of Lewis lung cancer in mice. The anti-tumor metastasis mechanism may be related to the effect on the expression of VEGF and nm23-H1 in Lewis lung cancer cells.
