二甲基甲酰胺(DMF)是一种用途广泛的有机溶剂,文献报道,DMF为亲肝性毒性,其作用详细机理尚未阐明。为探讨DMF对肝细胞钙稳态的影响及其与肝细胞损伤的关系,我们选用磷酸化酶a作为肝细胞浆钙离子浓度的间接指标,测定DMF与原代培养的大鼠肝细胞孵育一定时间后胞浆磷酸化酶a活性,同时测定GPT、GOT漏出量,并用台盼蓝拒染法观察细胞生长状况。
Dimethylformamide (DMF) is a widely used organic solvent. It has been reported in the literature that DMF is a hepatotropic toxicant. The detailed mechanism of its action has not been elucidated yet. In order to investigate the effect of DMF on the hepatocyte calcium homeostasis and its relationship with hepatocellular injury, we used phosphorylase a as an indirect indicator of the concentration of hepatocyte cytoplasm calcium and determined that the incubation of DMF with primary cultured rat hepatocytes The activity of cytoplasmic phosphorylase-a was detected after the time, and the GPT and GOT leakage were measured simultaneously. Cell growth was observed by trypan blue exclusion.
