目的:研究脑痛立停胶囊对偏头痛模型大鼠脑干及下丘脑原癌基因c—fos、c—jun基因表达的影响,探讨脑痛立停胶囊治疗偏头痛的作用机理。方法:建立电刺激上矢状窦(sSs)区硬脑膜大鼠偏头痛模型,给予脑痛立停胶囊灌胃治疗,以非甾体类抗炎镇痛药萘普生为对照药物。给药后4小时以免疫组化SABC法检测脑干和下丘脑c—fos、c—jun表达水平。计算机图像分析系统分析阳性细胞数、阳性细胞面积和阳性细胞灰度值。结果:电刺激SSS区硬脑膜偏头痛模型大鼠脑干和下丘脑c—fos、c—iun阳性细胞数及阳性细胞面积明显高于正常对照组和假手术组,经脑痛立停胶囊灌胃治疗后。模型大鼠脑干和下丘脑c—fos、c—jun阳性细胞数及阳性细胞面积明显下降,其中大剂量组作用明显,其作用强度与萘普生相当。结论:脑痛立停胶囊可能通过某些作用机制抑制脑干、下丘脑c—fos与c—jun基因表达,减弱或阻断了疼痛刺激在中枢神经的传导,发挥治疗偏头痛的作用。
Objective: To study the effect of Naotong Li-stop capsule on the expression of c-fos and c-jun genes in the brainstem and hypothalamus of rats with migraine and to explore the mechanism of Naotong Li-stop capsules in the treatment of migraine. METHODS: A migraine model of dural rats in the superior sagittal sinus (sSs) region was established by electrostimulation, and was given intragastrically by Naotong Li-stop Capsules. The non-steroidal anti-inflammatory analgesic naproxen was used as a control drug. The expression of c-fos and c-jun in the brain stem and hypothalamus was detected by immunohistochemical SABC method 4 hours after administration. The computer image analysis system analyzes the number of positive cells, the area of positive cells, and the gray value of positive cells. RESULTS: The number of c-fos and c-iun positive cells and positive cells in the brainstem and hypothalamus of rats with electrical stimulation of the dural migraine in the SSS area were significantly higher than those in the normal control group and the sham operation group. After stomach treatment. The number of c-fos and c-jun positive cells and the area of positive cells in the brain stem and hypothalamus of the model rats were significantly decreased, and the effect of the high-dose group was obvious, and its effect intensity was comparable to that of naproxen. Conclusion: Naotong Li-stop capsules may inhibit the expression of c-fos and c-jun genes in the brain stem and hypothalamus through certain mechanisms, weaken or block the transmission of painful stimuli in the central nervous system, and play a role in the treatment of migraine.
