目的:考察人参不同制备部位对小鼠脾淋巴细胞生物热活性影响的差异,为中药活性的筛选等多个方面的研究建立新的方法。方法:采用瑞典生产的2277型热活性检测系统,以对淋巴细胞的最大发热量,代谢热平均值及衰减速率常数为考察指标,进行研究;同时,采用MTT法对实验结果进行了验证。结果:ZG2对淋巴细胞的微量热的影响没有统计学意义,ZG3、ZG4、ZG5不同剂量均可使可使淋巴细胞的Pav、Pmax增加和kd降低而促进其代谢产热(与空白组比P<0.05),作用强弱次序为:ZG4>ZG5>ZG3,MTT法所得的实验结果与其基本一致。结论:应用生物热力学的方法可很好的检测脾淋巴细胞的生长代谢的微量热变化,为其在中药药效物质筛选等多个方面的研究提供了新的依据。
OBJECTIVE: To investigate the differences in the effects of different preparation sites of ginseng on the biothermic activity of spleen lymphocytes in mice, and to establish a new method for the study of the activity screening of traditional Chinese medicines. Methods: The Swedish 2277 thermal activity detection system was used to study the maximum calorific value of the lymphocytes, the mean value of metabolic heat, and the decay rate constant. The results of the experiment were verified by MTT method. Results: The effect of ZG2 on microcalorie of lymphocytes was not statistically significant. Different doses of ZG3, ZG4 and ZG5 could increase the Pav, Pmax and kd of lymphocytes and promote the metabolic heat production (Compared with the blank group P <0.05), The order of the effects is: ZG4> ZG5> ZG3, and the experimental results obtained by the MTT method are basically the same. Conclusion: The method of biothermodynamics can be used to detect the micro-thermal changes of spleen lymphocyte growth and metabolism, and provide a new basis for the research on the screening of pharmacodynamic substances in traditional Chinese medicine.
