为了对转基因抗虫玉米转育株Bt蛋白的表达特性进行分析,用Cry1Ab/Cry1Ac平板试剂盒定量检测了3种含Bt基因的玉米回交后代品系BC1F1(HZ3152、HZ3342、HC0142)在相同生长时期不同组织和不同生长时期相同组织中Bt蛋白的表达量。结果表明,在乳熟期BC1F1不同组织中Bt蛋白表达含量的高低顺序为:叶片>胚乳>花丝>种子苞叶;在BC1F1不同的生长发育阶段(苗期、抽丝期和乳熟期),叶片中Bt蛋白的含量随生育期的推移呈明显先上升后下降趋势,Bt蛋白含量在抽丝期达到最大,之后又开始下降;苗期(HC0142、HZ3152、HZ3342)Bt蛋白的含量(4.16、3.75和3.72μg·g-1)和乳熟期(5.56、4.58和4.79μg·g-1)的含量分别是抽丝期(6.05、5.36和5.50μg·g-1)的68%、69%、67%和91%、85%、87%。一般在玉米生长后期Bt蛋白的浓度有所下降,但幅度不大,对其抗性不会造成太大影响。该研究表明Bt蛋白在玉米回交后代中可以稳定表达,对Bt的相关研究为转基因育种等提供了参考依据。
In order to analyze the expression characteristics of Bt protein in transgenic insect-resistant maize plants, three BC1F1 (HZ3152, HZ3342, HC0142) lines with backcross of the Bt gene were quantitatively detected by Cry1Ab / Cry1Ac plate kit in the same growth period Bt protein expression levels in different tissues and in the same tissues at different growth stages. The results showed that the order of the expression of Bt protein in different tissues of BC1F1 was: leaf> endosperm> filament> seed bract; in the different growth and development stages of BC1F1 (seedling, spinning and milking) The content of Bt protein in leaves increased firstly and then decreased with the growth period. The content of Bt protein reached its maximum at the spinning stage and then decreased again. The content of Bt protein in seedling stage (4.16, 3.75 and 3.72 μg · g-1) and milky ripe stage (5.56, 4.58 and 4.79 μg · g-1) were 68% and 69% of spinning stage (6.05, 5.36 and 5.50 μg · g- , 67% and 91%, 85% and 87% respectively. Generally in late corn Bt protein concentration decreased, but the magnitude is not large, its resistance will not cause much impact. This study shows that Bt protein can be stably expressed in backcross progeny of maize, and the related research of Bt provides a reference for transgenic breeding.
