目的探讨自制载血卟啉单甲醚(HMME)的聚乳酸-羟基乙酸(PLGA)高分子微球HMME/PLGA对卵巢癌SKOV_3细胞的急性细胞毒作用及增殖能力的影响。方法实验分为5组:(Ⅰ)单纯细胞(对照组),(Ⅱ)超声+细胞,(Ⅲ)超声+PLGA+细胞,(Ⅳ)超声+HMME+细胞,(Ⅴ)超声+HMME/PLGA+细胞。超声辐照声强0.50 W/cm~2,频率1 MHz,Ⅳ组和Ⅴ组HMME浓度为20μg/ml。以CCK8法检验各组不同超声处理时间(10s、30s、60s、90s)对卵巢癌SKOV3细胞存活率的影响,并于上述实验基础上检测各组对SKOV3细胞12h、24h、36h、48h增殖能力的影响。结果急性细胞毒实验及抑制增殖实验显示:V组(US+HMME/PLGA+cell)较其余各组能明显降低SKOV_3细胞的存活率(P<0.05),抑制肿瘤细胞的增殖活性(P<0.05)。结论自制的HMME/PLGA高分子微球,对卵巢癌SKOV3细胞有明显的杀伤、抑制功能,有望应用于体内的卵巢癌治疗。
OBJECTIVE: To investigate the effect of self-made HMME / PLGA microspheres HMME / PLGA on the acute cytotoxicity and proliferation of ovarian cancer cell line SKOV_3. Methods The experiment was divided into five groups: (Ⅰ) simple cells (control group), (Ⅱ) ultrasound + cells, (Ⅲ) ultrasound + PLGA + cells, (Ⅳ) ultrasound + HMME + cells and (V) ultrasound + HMME / PLGA + cells. The intensity of ultrasonic irradiation was 0.50 W / cm ~ 2 and the frequency was 1 MHz. The concentration of HMME in group Ⅳ and group Ⅴ was 20 μg / ml. The effects of different sonication time (10s, 30s, 60s, 90s) on the survival rate of ovarian cancer SKOV3 cells were tested by CCK8 method. On the basis of above experiments, the proliferation of SKOV3 cells at 12h, 24h, 36h, 48h Impact. Results The results of acute cytotoxicity assay and proliferation inhibition test showed that the survival rate of SKOV_3 cells was significantly decreased (P <0.05) and the proliferation activity of tumor cells was inhibited by US + HMME / PLGA + cell compared with the other groups ). Conclusion Self-made HMME / PLGA polymer microspheres have obvious cytotoxicity and inhibitory activity on ovarian cancer SKOV3 cells, which is expected to be used in the treatment of ovarian cancer in vivo.
