目的观察细粒棘球蚴囊液及原头蚴对体外培养BABL/c小鼠脾细胞TGF-β表达的影响。方法取BABL/c小鼠脾细胞,制成细胞悬液,加入不同浓度囊液/原头蚴共培养72h,用ELISA检测上清液TGF-β含量;同时收集细胞并提取mRNA,用RT-PCR检测TGF-βmRNA的相对表达量。试验以PBS与脾细胞混合培养组为对照。结果 ELISA检测囊液组和原头蚴组TGF-β分泌均增加,与对照组相比差异有统计学意义(P<0.05);RT-PCR检测囊液组和原头蚴不同浓度组及对照组TGF-βmRNA的相对表达量,差异均有统计学意义(P<0.05),但囊液/原头蚴不同浓度组间差异无统计学意义(P>0.05)。结论原头蚴和囊液均能刺激体外培养小鼠脾细胞TGF-β表达增加,提示原头蚴和囊液中含有能促使宿主产生TGF-β的抗原物质,这些抗原物质在包虫病的免疫逃避中发挥一定作用。
Objective To observe the effects of Echinococcus granulosus and protoscolex on the expression of TGF-β in splenocytes of BABL / c mice in vitro. Methods Splenocytes of BABL / c mice were used to prepare cell suspension. The cells were cultured for 72 hours with different concentrations of cysts / Procesian cercariae. The contents of TGF-β in supernatant were detected by ELISA. Meanwhile, PCR detection of TGF-β mRNA relative expression. The experiment with PBS and spleen cells mixed culture group as a control. Results The levels of TGF-β in cytoplasm and progenitor were all increased in ELISA group compared with control group (P <0.05). The concentrations of TGF-β in cytoplasm and progenitor cells were significantly increased (P <0.05). However, there was no significant difference between the different concentrations of cystic fluid / Echinococcus granulosus (P> 0.05). Conclusion Both procollagen and cyst fluid can stimulate the expression of TGF-β in mouse spleen cells cultured in vitro, suggesting that the progenitor cells and the cystic fluid contain antigenic substances that can induce the host to produce TGF-β. These antigenic substances are highly expressed in echinococcosis Immune escape play a role.
