目的测定甘草次酸改性前后的表观油水分配系数,考察其水溶性改善情况。方法建立紫外分光光度法,采用摇瓶法(正辛醇-水体系)测定甘草次酸及聚乙二醇单甲醚改性甘草次酸在不同油水比例以及不同p H值下的表观油水分配系数(lg P)。结果甘草次酸在无水乙醇中的最大紫外吸收波长为248nm,线性回归方程为A=0.0233C-0.0008(n=6,r=0.9999),在6.0~36mg/L范围内,线性关系良好。改性后甘草次酸的表观油/水分配系数下降到-0.538~-0.767。结论难溶性中药有效成分甘草次酸经聚乙二醇单甲醚改性后在水中的溶解性显著提高。
Objective To determine the apparent oil-water partition coefficients of glycyrrhetinic acid before and after modification, and to investigate the water-soluble improvement. Methods UV spectrophotometry was used to determine the apparent water-oil ratio of glycyrrhetinic acid and polyethylene glycol monomethyl ether glycyrrhetinic acid at different oil-water ratios and different p H values by shaking flask method (n-octanol-water system) Distribution coefficient (lg P). Results The maximum UV absorption wavelength of glycyrrhetinic acid in absolute ethanol was 248 nm. The linear regression equation was A = 0.0233C-0.0008 (n = 6, r = 0.9999). The linear range was 6.036 mg / L. The apparent oil / water partition coefficients of glycyrrhetinic acid decreased to -0.538 ~ -0.767. Conclusion The solubility of glycyrrhetinic acid, an active ingredient of the insoluble traditional Chinese medicine, in water after the modification with polyethylene glycol monomethyl ether was significantly increased.
