目的 研究乙型肝炎病毒(HBV)前 C区热点变异对宿主细胞HLA—I分子表达的影响。方法 构建HBV前C区野毒株及A83、A83/A86变异株真核表达质粒,转染HePG2细胞,鉴定目的基因在宿主细胞中的生物活性,流式细胞术检测HLA—I分子的表达。结果 PCR和 ELISA法分析能检测到目的DNA片段和HBeAg,转染细胞表达HLA—I分子的平均荧光强度不同,野毒株为1.3,前C区变异后平均荧光强度增加,尤以A83变异表达增强显著(17.6),A83/A86为7.3。结论 前C区热点变异后宿主细胞HLA—I分子的表达为上调。
Objective To study the influence of pre-HBV hot spot mutation on the expression of host HLA-I molecules in Hepatitis B virus (HBV). Methods The eukaryotic expression plasmids of the pre-HBV C region and A83, A83 / A86 were constructed and transfected into HepG2 cells to identify the biological activity of the target gene in host cells. The expression of HLA-I molecules was detected by flow cytometry. Results The DNA fragments and HBeAg were detected by PCR and ELISA. The mean fluorescence intensity of HLA-I molecules in transfected cells was different. The average fluorescence intensity of wild-type strain was 1.3 and the average fluorescence intensity of the pre-C region was increased, especially A83 Significantly increased (17.6), A83 / A86 was 7.3. Conclusion The expression of HLA-I in host cells was up-regulated after the hot spots in the former C region were mutated.
