目的研究脱氧胆酸对结肠癌细胞株HT29中环氧合酶2(COX-2)转录后调控的影响。方法通过荧光定量PCR,荧光素酶报道基因技术检测脱氧胆酸对COX-2转录后调控的影响。结果脱氧胆酸可促进COX-2mRNA半衰期的延长,上调含COX-23’UTR的荧光素酶报道基因的表达。结论脱氧胆酸上调结肠癌细胞株HT29中COX-2转录后调控,其主要作用位点在COX-23’UTR上。
Objective To study the effect of deoxycholic acid on the post-transcriptional regulation of cyclooxygenase 2 (COX-2) in colon cancer cell line HT29. Methods Fluorescein quantitative PCR and luciferase reporter gene technology were used to detect the effect of deoxycholic acid on the post-transcriptional regulation of COX-2. Results Deoxycholic acid increased the half-life of COX-2 mRNA and up-regulated the expression of luciferase reporter gene containing COX-23’UTR. Conclusion Deoxycholic acid can up-regulate the transcription of COX-2 in colon cancer cell line HT29. The main site of action is COX-23’UTR.
