目的:检测焦虑情绪模型大鼠额区皮层γ-氨基丁酸(GABA)B受体两个亚基GABABR1,GABABR2和腺苷酸环化酶(AC)表达的变化,初步探讨白香丹胶囊对焦虑情绪的干预机制。方法:白香丹胶囊由芍药苷、香附挥发油和丹皮酚配伍组成。大鼠随机分为正常组、模型组、白香丹胶囊组(200 mg·kg-1)、巴氯芬组(8 mg·kg-1),采用“孤养加异种大鼠入侵”方法,大鼠孤养2周,然后异种大鼠居住入侵刺激2周,制备焦虑情绪大鼠模型。从居住入侵第2周开始,药物组按相应剂量ig给药7 d,每日1次。通过糖水偏好实验、旷场实验和高架十字迷宫实验进行模型评价,用免疫荧光技术检测各组大鼠额区皮层GABABR1,GABABR2和AC的表达变化。结果:与正常组比较,模型组糖水偏好系数降低,旷场实验得分增高(P<0.05),高架十字迷宫实验进入开放臂次数百分数(OE%)和开放臂停留时间的百分数(OT%)降低(P<0.05),额区皮层GABABR1,GABABR2,AC表达水平降低(P<0.05)。与模型组比较,白香丹胶囊组和巴氯芬组旷场实验得分降低(P<0.05),OE%和OT%值升高(P<0.05),额区皮层GABABR1,GABABR2,AC表达水平升高(P<0.05)。结论:大鼠额区皮层GABABR1,GABABR2,AC表达下调可能与焦虑情绪的产生有关,白香丹胶囊抗焦虑作用的中枢机制可能与其恢复额区皮层的GABABR的表达和功能有关。
AIM: To detect the expression of GABABR1, GABABR2 and adenylyl cyclase (AC) in two subunits of γ-aminobutyric acid (GABA) B receptor in frontal cortex of anxiety model rats, Anxiety of the intervention mechanism. Method: Bai Xiangdan capsules from paeoniflorin, volatile oil and paeonol compatibility. The rats were randomly divided into normal group, model group, Bai Xiang Dan capsule group (200 mg · kg -1) and baclofen group (8 mg · kg -1) Methods: The rats were solitary for 2 weeks and then invaded for 2 weeks to induce anxiety in rats. From the second week of residential invasion, the drug group was given ig for 7 days at the corresponding dose once a day. The model was evaluated by sweet water preference test, open-field test and elevated plus maze test. The expression of GABABR1, GABABR2 and AC in frontal cortex were detected by immunofluorescence technique. Results: Compared with the normal group, the preference coefficient of syrup in the model group decreased and the open field test score increased (P <0.05). The percentage of OE% and OT% of the open arms decreased (P <0.05). The expression of GABABR1, GABABR2 and AC in frontal cortex decreased (P <0.05). Compared with the model group, the open field test scores of Bai Xiang Dan Capsule group and Baclofen group decreased (P <0.05), OE% and OT% increased (P <0.05), and the levels of GABABR1, GABABR2 and AC in frontal cortex Increased (P <0.05). CONCLUSION: Down-regulation of GABABR1, GABABR2 and AC expression in the frontal cortex of rats may be related to the generation of anxiety. The central mechanism of Bai Xiangdan capsule’s anti-anxiety may be related to the expression and function of GABABR in the frontal cortex.
