目的 :探讨巴戟天寡糖 (MW 97)抗抑郁作用的可能机制。方法与结果 :MW 97或NGF与皮质酮 2×10 -4 mol·L-1共孵PC12细胞 48h ,可防止皮质酮所致的PC12神经细胞损伤。进一步运用RT PCR方法研究表明 ,MW 97(10 0mg·kg-1)及去甲丙米嗪 (10mg·kg-1)慢性ip 2 1d ,可使大鼠前脑皮层NGF、脑源性神经营养因子(BDNF)及海马BDNFmRNA表达升高。结论 :MW 97抗抑郁作用机理可能与神经细胞营养因子表达升高 ,从而对皮质酮诱导的神经元损伤产生保护作用有关。
Objective: To explore the possible mechanism of antidepressant effect of Morinda officinalis (MW 97). METHODS AND RESULTS: PC12 cells co-incubated with MW 97 or NGF and corticosterone 2×10 -4 mol·L -1 for 48 h prevented PC12 nerve cell damage caused by corticosterone. Further studies using RT-PCR showed that MW 97 (100 mg·kg-1) and norripiprazine (10 mg·kg-1) chronic ip 21d induced NGF and brain-derived neurotrophy in rat precerebral cortex. Factors (BDNF) and hippocampal BDNF mRNA expression increased. Conclusion : The mechanism of antidepressant effect of MW 97 may be related to the increased expression of neurotrophic factor, which may be related to the protective effect of corticosterone-induced neuronal injury.
