泼尼松对雄性大鼠骨代谢的影响

目的探讨泼尼松对雄性大鼠骨代谢过程的影响。方法20只3月龄SD雄性大鼠,随机分成正常对照组(A组)、泼尼松组(B组),A组灌生理盐水4 ml·kg-1·d-1,B组予4 mg·kg-1·d-1泼尼松灌胃,共90 d。实验结束后,处死全部大鼠,取尺骨和血清做生化指标测定。结果泼尼松组大鼠尺骨羟脯氨酸(Hyp)的含量无显著变化,骨钙、骨磷、骨锌的含量显著降低;血清钙显著升高,碱性磷酸酶含量降低。结论泼尼松通过抑制成骨细胞的活性,减少骨形成,增加骨吸收,导致骨质丢失。在骨质丢失的过程中,最早出现无机质的丢失,继而出现有机质的丢失。 Objective To investigate the effects of prednisone on bone metabolism in male rats. Methods Twenty male SD rats of 3 months old were randomly divided into normal control group (group A) and prednisone group (group B). Group A received saline 4 ml · kg-1 · d-1, group B received 4 mg · kg-1 · d-1 prednisone intragastrically for 90 d. After the experiment, all the rats were sacrificed and the ulna and serum were taken for biochemical determination. Results There was no significant change in hydroxyproline (Hyp) content in the prednisone group, while the contents of osteocalcin, osteoporosis and bone zinc were significantly decreased. The levels of serum calcium and alkaline phosphatase were also decreased. Conclusion Prednisone can reduce bone formation, increase bone resorption and lead to loss of bone mass by inhibiting the activity of osteoblasts. In the process of bone loss, the earliest inorganic loss, followed by the loss of organic matter.
