目的观察电离辐射对EL-4细胞survivin蛋白表达的影响及与P53表达的关系。方法采用流式细胞术(FCM)观察EL-4细胞接受4GyX射线照射后,survivin蛋白的表达、P53表达及细胞凋亡的变化。结果4GyX射线照射EL-4细胞,Survivin蛋白表达受抑制,在4h明显降低,与对照组相比,差异有统计学意义(P<0·001);照射组P53蛋白表达水平均高于对照组,在此时间点上与对照组相比,差异有统计学意义(P<0·01)。照射后4h细胞凋亡率显著升高,与对照组相比差异有统计学意义(P<0·001)。结论survivin与P53在细胞凋亡调控中发挥重要作用,survivin表达与P53表达水平相关。
Objective To observe the effect of ionizing radiation on the expression of survivin in EL-4 cells and its relationship with the expression of P53. Methods Flow cytometry (FCM) was used to observe the changes of survivin protein expression, P53 expression and apoptosis in EL-4 cells exposed to 4Gy X-rays. Results Compared with control group, the expression of Survivin protein in 4Gy X-ray-irradiated EL-4 cells was significantly decreased at 4h (P <0 · 001), and the expression of P53 protein in irradiated group was higher than that in control group , At this time point compared with the control group, the difference was statistically significant (P <0.01). 4h after irradiation, the apoptosis rate was significantly increased compared with the control group, the difference was statistically significant (P <0.001). Conclusion Survivin and P53 play an important role in the regulation of apoptosis. The expression of survivin is correlated with the expression of P53.
