大豆氮素固定量的测定有乙炔还原法、~(15)N“A”值法、检查有无根瘤着生等间接法和~(15)N_2直接测定法。其中~(15)N_2直接测定法,除可得到 N 素固定量外,还能追踪所固定 N 素的去向。一般认为,将~(15)N_2以一定时间(1—2周)供给根际,如能从~(15)N_2的吸收量测定 N 素固定量的话,这个数值不仅表示日变化情况,也表示在供给~(15)N 期间随着环境的变化根瘤菌活性变化的真正固定量。因此~(15)N_2直接测定方法被认为是有很高使用价值的手段。
