目的探讨东海海参酶解液对小鼠空间和时间记忆的影响及其抗疲劳的功能。方法选用复合蛋白酶,在55℃的条件下酶解150min制得海参酶解液,灌胃ICR小鼠,42d后,利用Morris水迷宫、Y字形迷宫和转棒式疲劳仪进行空间、时间记忆能力和抗疲劳的研究;测定小鼠血清中超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活力以及丙二醛(MDA)的含量研究其抗氧化活性,利用荧光定量PCR研究记忆相关基因的表达。结果东海海参酶解液能显著强化小鼠的空间和时间记忆力,增强小鼠抗疲劳能力,提高血清SOD和GSH-Px的活性,并能降低血清MDA的含量,明显提高抗氧化活性。能够上调小鼠脑内记忆相关基因转录水平的表达。结论东海海参营养丰富,具有抗疲劳、抗氧化和提高记忆力的功能,可能是一种很好的保健食品原料。
Objective To investigate the effects of sea cucumber hydrolyzate on the spatial and temporal memory of mice and its anti-fatigue function. Methods The compound protease was used to enzymolysis for 150min at 55 ℃ to prepare the sea cucumber hydrolyzate. The ICR mice were given gavage. After 42 days, Morris water maze, Y-shaped maze and rod-like device were used for spatial and temporal memory And anti-fatigue. The activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) and the content of malondialdehyde (MDA) Fluorescence quantitative PCR was used to study the expression of memory related genes. Results Sea cucumber sea water enzymatic hydrolyzate significantly enhanced the spatial and temporal memory of mice, increased the anti-fatigue ability of mice, increased the activity of serum SOD and GSH-Px, decreased the content of serum MDA and significantly improved the anti-oxidative activity. Can upregulate the transcriptional level of memory-related genes in mouse brain. Conclusion The sea cucumber in the East China Sea is rich in nutrients and has the functions of resisting fatigue, antioxidizing and improving memory, which may be a good raw material for health food.
